Abstract
Effect of culture media and plant growth regulators on growth and development of proto-corm like bodies (PLBs) and subsequent plantlet regeneration of Vanda denisoniana were studied. The results showed that Vz MS (Murashige and Skoog) medium with 3% sucrose, 15% coconut water, 0.2% activated charcoal, 0.75% agar and 1 mg/L NAA gave the highest percentage of PLB multiplication at 95.83%. The highest number of shoot formation (16.33 shoots/explant) was obtained on Vi MS medium supplemented with 1 mg/L NAA in combination with 0.5 mg/L BA. The shoot cultured on Vi MS medium without plant growth regulators for 2 months, giving the highest length of shoots at 2.77 cm, 6 roots/plant and root length at 2.1 cm, with significant difference with vw (Vacin and Went) and NDM (New Dogashima Medium). However, there are no significant differences in term of number of leaves and leaf length. The complete plantlets had 90% รนTVival rate at 1 month after deflasking.
