This study was carried out in order to isolate and characterize gliding bacteria by collecting gliding bacteria by collecting 50 specimens for example, seawater, algae, soil, biofilm , rock, plastic and decaying material from coastal of Bandon estuary, Surat Thani province. Thirty-five isolate of the gliding bacteria were identified and classified into 5 groups according to their morphology, motility and Gram ร stain. Ten isolate were cultivated in SAP2 medium for 7 days and the supernatant of culture broth was precipitate by cold ethanol 95%. The results showed that the crude biopolymer from G2/9 gave the highest yield of 1.5 g/l. The SAP2 medium containing 10 g/l glucose give the highest yield of 2.6 g/l which was slightly higher than the obtained from using the original medium (1.73 times)
The biopolymer was partially purified and studied for its composition and properties. It was found to be water-soluble and acidic polymer containing and 32.83 % (พ/พ) total sugar 11.76% (พ/พ) protein. Fourier transform infrared (FT-IR) analysis showed that the biopolymer consisted of carboxyl, hydroxyl, amino and sugar derivative groups. The results showed that the crude biopolymer indicated that this gliding bacteria is highly potential for further investigation and development in absorb metal or biopolymer bead for encapsulate drug.
